Amy Winehouse nasce a Enfield, Middlesex, Inghilterra, in una famiglia ebraica di padre farmacista e madre infermiera. Cresce a Southgate (Londra), dove frequenta la Ashmole School. All’età di dieci anni fonda un gruppo rap amatoriale chiamato Sweet ‘n’ Sour, as Sour. Lo descrive come la versione bianca e ebraica delle Salt ‘n’ Pepa. A dodici anni frequenta la Sylvia Young Theatre School, ma a tredici viene espulsa perché non si applica e a causa di un piercing al naso. In seguito frequenta la BRIT School a Selhurst, Croydon.
Cresce ascoltando diversi tipi di musica (dalle Salt ‘n’ Pepa a Sarah Vaughan) e riceve la sua prima chitarra a tredici anni. Inizia a cantare come professionista all’età di sedici anni, dopo che la sua amica e cantante soul Tyler James manda una sua demo ad un talent scout. Firma per la sua etichetta discografica attuale, la Island/Universal.
